Mixing procedure and incubation time play a major role in kinetic solubility determinations. A method based on ltration in 96-well plates followed by UV analysis has been developed by pION Inc. and marketed under the m-sol evolution instrument (www.pion-inc.com). Basically aliquots of a 10-mM solution in DMSO are mixed with buffer, incubated, and ltered. The UV spectrum of the ltrate is then compared with a reference standard to quantify the amount of substance in solution. Respective shapes of the ltrate and the reference spectra are also analyzed in an attempt to determine whether an impurity has interfered with the assay. For ionizable molecules algorithms were also derived to account for the presence of DMSO.

This method is particularly well suited for the analysis of ionizable molecules, for which solubility is in uenced by pH (70 80% of generic drugs). It does not require the development of any analytical method but the ionization constants have to be determined relatively accurately prior to the analysis. The main advantages of this approach are (1) there is no need to separate the solute from the precipitate, (2) the pH is automatically adjusted by the addition of KOH/HCl, therefore no buffer is needed, (3) the equilibrium kinetics are taken into account by the system using a modi ed version of the Noyes-Whitney equation, (4) the Noyes Whitney equation gives the solubility pH-pro le, and (5) it covers an exceptional dynamic range (from mg/mL down to ng/mL). There are, however, a number of limitations inherent to the technology: (1) it is only applicable to ionizable molecules, (2) weak acids (pKa > 11) and weak bases (pKa < 3) cannot be measured because the ionization is then shifted to a range outside the dynamic range of the pH electrode, and (3) it is not a high-throughput method (1 2 compounds/day) Saturation Shake-Flask This method is used as a complementary tool for neutral compounds or for weak acids or bases. Its main advantage is that ionizable as well as neutral compounds can be analyzed. For the former, however, one has to assume that the components of the buffer do not in uence the solubility, particularly when one performs a measurement at a pH value where the molecule is strongly ionized. Prior to each analysis one needs to establish an analytical method and to build a calibration curve. In most cases it can be achieved by reversephase HPLC with a C18 or C8 column. The main dif culty of the method lies in the separation of the phases where decantation by centrifugation or ltration is used. In each case, there is some risk that the solubility is affected by this phase-separation step. When decantation is used the solubility is potentially overestimated, while the opposite is true when ltration is used. In today s drug discovery environment one usually prefers to separate the phases by decantation through centrifugation because the large majority of the research compounds are rather hydrophobic (CLOGP > 3).
