2. Hepatocellular adenomas and carcinomas: Hepatocellular neoplasms were induced in both sexes of rats and mice at furan low-dose levels of 2 and 8 mg/kg bw, respectively (Table 2.4.8). The increase in incidence was dose-related. Hepatocellular neoplasms were not observed at the 9- or 15-month interim evaluation in rats. Background (control mice) incidence of hepatocellular adenoma/carcinoma in male mice was high (52%). Hepatocellular carcinoma was rst observed at the 15-month interim evaluation in 2 of the 10 male rats dosed with 30 mg/kg for 13 weeks of a stop-exposure study. 3. The NTP studies demonstrated that furan induced cytotoxic and carcinogenic effects at all doses tested in rats and mice, and a no observed adverse effect level (NOAEL) was not identi ed. A recent study by Moser et al. (G. Moser, 2007, personal communication) examined the hypothesis that cytotoxicity and cell proliferation may play a role in the liver tumor formation in female mice treated with furan. Like the NTP bioassay, female B6C3F1 mice were exposed to furan, 5 days a week, by gavage in corn oil for 3 weeks or 2 years. Furan dosages included the carcinogenic doses used in the NTP bioassay and lower dosages of 0, 0.5, 1.0, 2.0, 4.0, and 8.0 mg/kg bw. There was a dose response relationship between the incidence of liver tumors and the induction of cytotoxicity and cell proliferation. The thresholds (Lowest Observed Adverse Effect Level, LOAEL) for cytotoxicity and tumors were 1.0 and 4.0 mg/kg bw, respectively. An increase in cell proliferation in the 3-week study was signi cant only at the 8.0 mg/kg bw dosage. This study did identify the NOAELs for both cytotoxicity and hepatocarcinogenicity in female B6C3F1 mice. Furthermore, it demonstrated that furan-induced liver tumorigenicity is a threshold-dependent event, which occurs at doses ( 4.0 mg/kg bw) well above the cytotoxic levels ( 1.0 mg/kg bw). The most sensitive end point and species/sex combination was the incidence of cholangiocarcinomas (bile duct tumors) in female F344 rats (Table 2.4.7). In this bioassay, there were four dose groups: a control, 2 mg/kg/day, 4 mg/kg/day, and 8 mg/kg/day. The incidence of cholangiocarcinoma was 0% in 50 controls and near 100% in all other dose groups. In the NTP study, interim sacri ces were conducted at both 9 and 15 months. The data at 15 months showed a saturated dose response similar to that seen at 24 months (lifetime).
